研究方向

以假单胞菌、芽孢杆菌、大肠杆菌和谷氨酸棒杆菌等原核细菌和酵母菌为对象,主要进行环境降解、底盘细胞改造、功能元件研发与适配性研究、基因表达活性调控、作用机制与应用技术基础等方面的研究工作。目前主要科研方向

1)假单胞菌降解环境难降解和有毒化合物的功能基因、降解活性、作用途径与机制;

2)运用CRISPR-Cas9/CRISPR-Cas12f1CRISPR-RNA等基因编辑工具进行假单胞菌、芽孢杆菌、大肠杆菌和谷氨酸棒杆菌基因组简约编辑,规模性删除基因组冗余基因和改造细菌底盘细胞;

3)体内连续进化并联合体外进化策略,以理性设计、半理性设计和构建自组装人工多酶复合体等生物技术策略实现对不同酶蛋白的分子改造,大幅提升酶催化活性与酶底物适配性

4)针对细菌和酵母细胞工厂不同元件的适配性研究。已在假单胞菌、大肠杆菌和酵母菌成功发掘不同强度的系列启动子、RBS、群体感应系统单元和菌体自杀系统单元,以适应于不同使用目的需要。

研究成果

先后主持国家863项目1项、国家973项目课题1项、国家自然科学基金重点项目课题2项和国家自然科学基金面上项目7项,以及主持省部级和横向各类项目20余项。发表SCI论文80余篇,其它学术论文30余篇,参与撰写英文学术专著2部,中文学术专著2部,获的授权国家发明专利7项。参与获得湖北省技术发明一等奖、农业部科技进步三等奖和武汉市科技进步二等奖等奖励。
